ABSTRACT

This study describes the organic synthesis of 5-[2-amimo-4-styryl pyrimidine-4-yl]-4-methoxy benzofuran-6-ol [SPBF] as an example of a benzofuran derivative used as a new series of amyloid imaging agents. These benzofuran derivatives may be useful amyloid imaging agents for detecting B-amyloid plagues in the brain of Alzheimer's disease. The precursor is 1-[6-hydroxy-4-methoxybenzofuran-5-yl]-phenyl butadiene ketone, which react with guanidine hydrochloride. The purification process was done via crystallization using solvent ethanol. The overall yield was 75% and the structure of the synthesized compound was confirmed by correct analytical and spectral data. Also, The synthesized compound was labeled with radioactive iodine -125 via electrophilic substitution reaction, in the presence of iodogen as an oxidizing agent, the labeling process was carried out at 95°C for 20min. The radiochemical yield was determined by using a thin layer chromatography and the yield was equal to 80%. Preliminary an in-vivo study examined normal mice after intravenous injection through the tail vein and the data showed the labeling compound was quickly cleared from most body organs. The radioiodinated compound showed high brain uptake. The results of this study suggest that radioiodinated [SPBF] may be useful as a brain imaging agents

ABSTRACT

The distribution of creatinine, one of the toxic guanidine compounds, in various tissues has not been studied in detail by using radiolabeled creatinine. Our objective was to investigate the biodistribution of creatinine labeled with 99m technetium (99mTc) by the stannous (II) chloride method in healthy male Wistar rats. Quality controls were carried out by radio thin layer chromatography, high-performance liquid chromatography, and paper electrophoresis. The labeling yield was 85 ± 2 percent under optimum conditions (pH 7 and 100 æg stannous chloride). Rats (N = 12) were injected intravenously with 99mTc-creatinine and their blood and visceral organs were evaluated for 99mTc-creatinine uptake as percent of the injected dose per gram wet weight of each tissue ( percentID/g). The lowest amount of uptake was detected in the brain and testis. When the rate of uptake was evaluated, only the kidney showed increasing rates of uptake of 99mTc-creatinine throughout the study. Kidneys showed the highest amount of uptake throughout the study (P < 0.001 compared to all other organs), followed by liver, spleen and lung tissue.

ABSTRACT

In the present investigation egg capsules of Gyraulus convexiusculus were treated with different concentrations of baygon. A dose and duration dependent deviations in the number of negatively charged protein fractions in the trochophore and veliger larval stages were observed. It resulted into anomalies in the morphogenesis and organogenesis of corresponding larval stages. Most of the protein bands showed the decrease in the protein positive intensities in comparison to control. This suggested that baygon causes larval toxicity in Gyraulus convexiusculus.

ABSTRACT

Administration of glucocorticoids one of the corticosteroid hormones, is one of the most important therapeutic strategies in ophthalmologic treatment for various ocular diseases. The most principle in ophthalmologic use of corticosteroids is producing less severe effects or complications, obtaining the most efficacious, with long duration. Evaluation of the effect of corticosteroid drops on the intraocular pressure, aqueous humor, vitreous humor proteins and lipids. New Zealand males' rabbits were subjected to local administration of Dexamethasone 0.1% drops for two period's times. The intraocular pressure was measured and after decapitation of the animals, the aqueous and vitreous were withdrawn and analyzed with different techniques for protein analysis. Then lipids were extracted, purified and extensively studied using different techniques. The results indicated an increase in the intraocular pressure, decrease in protein content, also the chromatographic patterns and the electrophoresis mobility were changed compared to normal. Moreover, the profile of vitreous lipids had the same characteristics as the normal and associated with fluctuated changes in its proteins and lipids component. The treatment of the inflamed eyes with Dexamethasone 0.1% indicating a change in the protein concentration, in the molecular weight of aqueous humor protein molecules and resulted in an increase in intraocular pressure which indicated an induction of glaucoma. Also long term administration of Dexamethasone 0.1% induced a change in vitreous lipids and proteins

ABSTRACT

Eales disease is an obliterative vasculopathy that usually involve peripheral retina of young adults. It is thought to be a nonspecific tissue response to a number of agents and exact etiology remains unknown. Many diseases reflect some alterations in electrophoretic pattern of serum proteins. This study was carried out to investigate electrophoretic pattern of serum proteins in Eales disease so as to throw some light on possible etiological factors in these patients. Thirty-two patients of Eales disease constituted the subject material for this study. Paper electrophoresis of the sera was performed by cellulose acetate method on Whatman filter paper No.10 using barbitone buffer of pH 8.6. Both patients and controls underwent complete ocular and clinical examination and followed up for one year. This was a descriptive study. Ten patients [30%] had raised total serum proteins out of 32 patients and ten patients [30%] had raised gamma globulin. Serum albumin was decreased in seven patients [20%] out of thirty-two patients of Eales disease. In our study 30% increase in total serum proteins and gamma globulins in Eales disease patients points to a possible role of some immunological process which manifests as retinal perivasculitis in eyes. An increase in erythrocyte sedimentation rate in >85% of cases is consistent with this suggestion

ABSTRACT

The difficulty of a reliable diagnosis of pancreatic diseases by scintiscanning, is mainly derived from the lack of adequate radiopharmaceuticals. With this purpose (125) I-L(-3) Iodo-a-Methyl Tyrosine ((125) I-IMT) has been studied, which has also been used for the diagnosis of different kind of brain tumors. The purpose of this work is the development of a quick and easy method for the synthesis and purification of the (125) I-IMT in order to be used in a Nuclear Medicine Service. The L-(-Methly Tyrosine was labeled with (125) I using I-/I03 and afterwards purified by an aniomic exchange resin. The labeling yield obtained was (96.0+0.5) per cent when the incubation time was 15 minutes. No significant statistical differences were observed when the incubation time was extended to 1 hour. Biodistribution studies in mice show that the percentage of activity concentration in pancreas is (34.24+14.03) per cent at 15 minutes post injection, remaining constant for 30 minutes. The pancreas/liver ratio 15 minutes after the injection of the labeled product was (12.22+3.59) and it remained constant for 45 minutes more. These results show that (125) I-IMT cab be used as a diagnostic agent for pancreatic diseases. Since (123) I was not avilable at the moment, this new methodology was developed with (125) I.

ABSTRACT

Serum protein electrophoresis done on 1100 patients with various diseases in one year demonstrated M-band in 31 patients. Most (87%) had the classical features of plasma cell dyscrasia (PCD), however a few had unusual presentations which are highlighted. A 22-year-old male operated for a massive tumour of the scapula clinically diagnosed as chondrosarcoma revealed plasmacytoma with amyloid on histology. Another case of kala-azar presented with features akin to that of PCD and one case had dual malignancies. Such a high incidence of PCDs with varied picture in a short time is not usually seen in other parts of this country; a fact which may be due to lack of awareness.

El estudio de las alteraciones del metabolismo de los aminoacidos urinarios ha motivado el desarrollo de dos tecnicas simples, rapidas y economicas, para el uso en Quimica clinica. Se describe la celda electroforetica disenada, asi como todas las operaciones requeridas para lograr dicho objetivo (tratamiento de los soportes usados y de las muestras, buffers, condiciones de corrida, distintos metodos y reactivos de revelado, desintometria). Se comparan los resultados obtenidos para los soportes empleados, papel y acetato de celulosa gelatinizado, usando para ambos igual sistema de refrigeracion y los resultados obtenidos en comparacion con otra forma de enfriamiento del sistema, utilizada previamente. Tambien se discute la aplicacion de mayores voltajes y la importancia de la estandarizacion en la coloracion. Trabajando con acetato de celulosa gelatinizado se pueden valorar por densitometria las corridas, lo cual brinda una util herramienta a los laboratorios clinicos

ABSTRACT

We report a case of generalized plane xanthoma which showed no-rmolipoproteinemia and was not associated with unerlying disorders or other type of xanthomas. A 48-year-old female has noticed well defined, slightly elevated, yellowish plaques which occurred on the periorbital areas, sides of the neck, scapular areas, anterior chest, buttocks and lower extremities for 3 years. Biopsy specimen taken from the neck showed perivascular accumulations of foam cells in the upper and mid dermis. Paper electrophoresis didn't, show elevation of any lipoprotein.

As vitaminas A (retino), B1 (tiamina), B6 (piridoxina), B12 (cianocobalamina), C (ácidoo ascórbico) e E (alfa-tocoferol), foram analisadas por eletroforese inidimensional em papel (qualitativo Klabin), usando como eletrólito soluçöes aquosas de carbonato de amônio 0,050, 0,075 e 0,100 M, sob um gradiente de potencial de 12,6V/cm. As melhores separaçöes ocorreram com o eletrólito nas concentraçöes 0,050 M (30 e 45 minutos) e 0,100 (45 e 60 minutos), quando 05 (cinco) vitaminas foram perfeitamente separadas: A (ou B6)-B1-B12-C e E

Os antibióticos fosfomicina, rifamicina, dicloxacilina penicilina G benzatina e gentamicina foram analisados por eletroforese unidimensional em papel (qualitativo Klabin), usando como eletrólito soluçöes aquosas de carbonato de amônio de concentraçöes 0,08, 0,10, 0,30 e 0,50 M, sob um gradiente de potencial de 12,6 V/cm. Todos os antibióticos citados apresentaram-se bem separados, utilizando o eletrólito nas concentraçöes 0,19 M (25 minutos) e 0,50 M (15 minutos)

ABSTRACT

Forty pregnant females of third trimester and their twenty non-pregnant controls were screened for Hb%, PCV and serum protein differentials in relation to parity. Paper electrophoresis method revealed significant rise in mean levels of serum alpha 1, alpha 2 and beta-globulins in pregnant females as compared to non-pregnant controls, which may be due to the rise in pregnancy-associated-proteins. Radial immunodiffusion method revealed rise in mean serum IgM level with increasing parity, whereas a decrease in mean serum IgG level was observed which might be due to the placental membrane-transfer and/or decreased IgG synthesis. Haematological investigations revealed decreased Hb% and PCV which may have been caused both due to increased demand of nutrients as well as by haemodilution, associated with water retention during pregnancy.

ABSTRACT

We report an atypieal case of xanthoma without evidence of underlying disease, A 50-year-old asymptomatic, yellowish, flat plaques on the upper eyelids, trunk and thighs, and huge, deep-seated tumors on anterior aspect of the neck, back and ankle areas of 3 years duration. Biopsies were performed at the lesions of the neck and hack. Two biopsy specimens showed the same findings of xanthoma, respectively. The appearance of refrigerated serum was clear. Paper electrophoresis didn't show elevation of any lipoprotein, suggesting normolipoproteinemia.

ABSTRACT

A 22-year-old male has had multiple, grouped, asymptomatic, yellowish flat papules, nodules, and plaques on the knees, elbows, butteeks, hands, and feet. Yellowish nodules first appeared on the heels after birth, and gradually increased in size and number and spread to the knees, elbows, buttocks, and hands. Physical examination revealed normaI except for the skin lesions. Gross finding of the serum was clear and paper electrophoresis revealed marked increase in betalipo-protein and mild increase in prebetalipoprotein, suggesting type II hyperlipoproteinemia Serum cholesterol was 510 mg%, and serum triglyceride was 190mg%. Histopathology showed many aggregates of foam cells on H-E stain and many lipid droplets in the dermis on oil red O stain.

ABSTRACT

A 43 year-old Korean male has had multiple, grouped, asymptomatic, yellowish brown flat papules and nodules on the knees, elbows, buttocks, palms and soles for 3 years. Small yeIlowish, discrete papules first appeared on the right palm and left elbow, and gradually increased in size and number and spread to knees, buttocks and soIes. Physical examination revealed normal except for the skin lesions and no familial occurrence was noted. Gross finding of the serum was turbid, more or less creamy and paper electrophoresis revealed marked increase of prebetalipoprotein, increase of betalipoprotein and alphalipoprotein and absence of chylomicron, suggesting type III or type IV hyperlipoproteinemia. Serem choleterol was 200mg% and fasting blood sugar was 115.0mg%. with normal glucose tolerance test. Histopathology showed many aggregates of foam cells on H-E stain and many lipid droplets in the dermis on Sudan red stain. Based on the clinical and histopathological findings and lipoprotein analysis by the paper electrophoresis, the patient was diagnosed as type IV hyperlipoproteinemia with xanthoma tuberosum.
